Abstract

Kadar kafein sesuai peraturan Kepala Badan POM tidak boleh melebihi 150 mg per hari. Penetapan kadar kafein dalam Farmakope Indonesia edisi V mempersyaratkan menggunakan metode Kromatografi Cair Kinerja Tinggi. Oleh karena itu Balai POM di Serang perlu melakukan uji verifikasi terhadap metode tersebut agar memenuhi persyaratan pengujian sesuai dengan kebutuhan. Tujuan penelitian ini adalah melakukan optimalisasi penetapan kadar kafein dalam serbuk minuman energi secara kromatografi cair kinerja tinggi dan untuk mengetahui kadar kafein dalam sampel serbuk minuman energi yang dijual di Kota Serang. Penetapan kadar kafein dilakukan menggunakan kromatografi cair kinerja tinggi dengan kolom : Oktadesilsilan (4,6 mm x 250 mm, ukuran partikel 5 µm), fase gerak : Air-Metanol-Asam asetat glasial (69:28:3), laju alir : 1,0 mL/menit, laju penyuntikan 20 µL, detektor : UV pada panjang gelombang 275 nm. Dilakukan verifikasi metode analisis dengan hasil Uji Selektivitas memenuhi persyaratan, Uji Presisi dengan RSD 3,827% (syarat 3,7-5,3 %), Uji Akurasi rata-rata perolehan kembali 98,80 % (syarat 90-107%), uji Linearitas dengan Vx0=0,950 (syarat < 2,0) dan r=0,999678 (syarat0,995). Hasil pengujian sampel menunjukkan bahwa sampel pertama hingga sampel keempat tidak memenuhi persyaratan batas maksimum kafein dalam makanan dan minuman, sedangkan sampel kelima memenuhi persyaratan dengan kadar dibawah 50 mg.
